- 方法通過(guò)RT PCR方法 ,從MT2細胞系克隆CT LA4全長(cháng)基因 ; 經(jīng)引物重疊和半巢式PCR改造 ,獲得含抑瘤素M信號肽的CTLA4胞外段DNA ,然后克隆至真核表達載體pIg ,轉染COS和CHO K細胞表達。Methods The full gene of the CTLA4 was amplified from MT2 cell lines by RT PCR, reformed with an overlapping primers and nested primers, then cloned into the expression vector pIg, consequently, the CTLA4 Ig was expressed in the COS and CHO K after transfection with pIg CTLA4. Results Sequencing data indicated that the CTLA4 gene was exactly identical to the sequence in the GenBank.
